Microfluidic Platform with Circular Microchannels for Facile Cell Trapping and Single Cell Analysis

A simple method to fabricate circular microchannels in polydimethylsiloxane (PDMS) is presented and used for facil single cell trapping at low vacuums. A coating of liquid PDMS is applied on the walls of square microchannels, to give them a circular cross section. Diameters ranging from few micrometers to few hundreds of micrometers were achieved. Different coating conditions were systematically tested. Design curves are reported for one to choose appropriate coating conditions for obtaining a desired diameter. A cell trapping device with 24 circular channels was used to immobilize 24 SiHa cells (cervical cancer cell line) at vacuum pressures of only -100 Pa.
